Assay Description: We tested the binding affinity of 6 small molecule JAK inhibitors (Alvocidib, Nintedanib, Bosutinib, Tofacitinib, Baricitinib, and S-Ruxolitinib) for JAK and other kinases (JAK1, JAK2, JAK3, TYK2 and DCLK3).
Assay Protocol: Compounds were sent to Eurofin (DicoverX/Ambit) and binding was tested according to the KdELECT(R) protocol. The KdELECT(R) assay platform is based on a competition binding assay that is run for a compound of interest against a selected panel of kinases. The assay has three components: a kinase-tagged phage, a test compound, and an immobilized ligand that the compound competes with to displace the kinase. The amount of kinase bound to the immobilized ligand is determined using quantitative PCR of the DNA tag. Results for each kinase are reported as "Kd".
