Systematic analysis of BRAFV600E melanomas reveals a role for JNK/c-Jun pathway in adaptive resistance to drug-induced apoptosis

The following data browsing tool allows you to explore across the full panel of cell lines studied the relative contribution to our partial least squares regression (PLSR) model of each molecular variable analyzed. Two versions of the PLSR loading plots are shown: one that is derived from models including all 21 molecular signals (top) and one that is derived from models including all molecular signals except pMEK and pERK (bottom).

This browser displays paired scatterplots for each of the cell lines studied, similar to those shown in Figure 1E of Fallahi-Sichani et al. (The data shown in this figure is the same as appears in the top two scatterplots displayed when this page first loads.)

Each scatterplot shows the relationship between the first and second PLSR model loadings from an analysis of the covariation of molecular signals with cell viability and apoptosis fraction in BRAFV600E/D melanoma cell lines.

Each plotted point corresponds either to an RPPA measurement/time point combination or to viability.

The first and second scatterplots in the pair shown for each cell line correspond, respectively, to analyses in which pMEK and pERK were included or excluded. Hence, the second scatterplot of each pair never contains points for pMEK or pERK. Furthermore, data is not available for some combinations of factors.

The corresponding axes over all scatterplots have the same range.

Interactions

  • Clicking on a cell line on the Cell_line panel...
    • adds it to the set of selected cell lines; and
    • and brings up the scatterplot pair for that cell line.
  • Hovering the cursor over a measurement in the RPPA_measurement panel...
    • highlights the datapoints in the scatterplots that correspond to it.
  • Hovering the cursor over a scatterplot...
    • ...causes its plot coordinates to be displayed in the info strip along the top of the scatterplots;
    • if, in addition, the cursor is over some datapoint P, ...
      • the metadata for P is also displayed in the info strip;
      • the measurement MP that P corresponds to will be highlighted in the RPPA_measurement panel; and
      • datapoint P, and all the other datapoints in the scatterplots corresponding to the same measurement MP, will also be highlighted.
  • Clicking on a measurement on the RPPA_measurement panel...
    • ...adds it to the set of selected measurements; and
    • fixes the highlighting for that measurement and all the associated datapoints.
  • Clicking on the RPPA_measurement panel's reset button clears the set of selected measurements.
  • Clicking on the Cell_line panel's reset button fully resets the browser.
